چکیده

Fine interconnections or "cross-bridges" have been observed between cytoplasmic microtubules in many different organisms (1, 3, 7, 9, 10, 12, 13) . Recently, similar bridges have been noted between the microtubules of the spindle apparatus in dividing plant and animal cells (4, 6, 7, 11, 14) . Increasing attention is being directed toward intertubule bridges, since they may play an important role in microtubule-mediated motility (1, 8, 9) . In this brief note we present our observations on the structure of arms and bridges on the microtubules of the spindle apparatus in cultured human cells (HeLa and WI-38) and in the endosperm of the African blood lily, Haemanthus katherinae . We demonstrate the presence of bridges between kinetochore tubules, between continuous tubules, and, during later stages of mitosis, between the tubules of the interzone . We also show bridges between tubules and vesicles in the postmitotic stem of HeLa cells and in the phragmoplast of Haemanthus .

The arrangement of microtubules in serially sectioned spindles of the alga Cryptomonas.

Serial sections of mitotic spindles of the marine cryptophycean alga, Cryptomonas, were analysed to determine what types of microtubules they contained and which of these microtubules came close enough to each other (50 nm or less) for the commonly described crossbridging to be possible. Interpolar microtubules were rare (less than or equal to 1%) but from prometaphase through anaphase there wa...

A TACC3/ch-TOG/clathrin complex stabilises kinetochore fibres by inter-microtubule bridging.

Kinetochore fibres (K-fibres) of the spindle apparatus move chromosomes during mitosis. These fibres are discrete bundles of parallel microtubules (MTs) that are crosslinked by inter-MT 'bridges' that are thought to improve fibre stability during chromosomal movement. The identity of these bridges is unknown. Clathrin is a multimeric protein that has been shown to stabilise K-fibres during earl...

Mechanisms and Regulation of the Mitotic Inheritance of the Golgi Complex

In mammalian cells, the Golgi complex is structured in the form of a continuous membranous system composed of stacks connected by tubular bridges: the "Golgi ribbon." At the onset of mitosis, the Golgi complex undergoes a multi-step fragmentation process that is required for its correct partition into the dividing cells. Importantly, inhibition of Golgi disassembly results in cell-cycle arrest ...

Arms and Bridges on Microtubules in the Mitotic Apparatus

Except for reported differences in diameters, the findings on fine structure of microtubules (MT) involved in mitosis and meiosis are consistent throughout the literature. Whatever the sources of cells for study, plant (14, 10, 17, 6) or animal (14, 9, 13, 20), no deviations have turned up, other than in centrioles which have tubules arrayed in triplets. Exceptions to the MT structure of the mi...

Chromosomal instability and cytoskeletal defects in oral cancer cells.

Oral squamous cell carcinomas are characterized by complex, often near-triploid karyotypes with structural and numerical variations superimposed on the initial clonal chromosomal alterations. We used immunohistochemistry combined with classical cytogenetic analysis and spectral karyotyping to investigate the chromosomal segregation defects in cultured oral squamous cell carcinoma cells. During ...
